Before you start this program, please make sure that:

  1. You have finished or nearly completely finished the backbone and sidechain assignments;
  2. You have already opened the necessary datasets and loaded their PeakLists as well as chemical shift assignments into the NMRVIEW (Drawing datasets is not necessary)

1. Setting your preferences

PeakList & Dataset: You are recommended to select the PeakLists firstly and their associated datasets will be automatically displayed. If they are not correct, you can manually key in the  correct one. If you don't have one of those spectra, please select or key in "NONE".

Dimension: It is similar to that in NMRView Attributes of spectrum window. You are only allowed to choose its attribute. "H" in 2D experiment or "HN/HC1" in 3D experiment corresponds to direct dimension (F1). "H2", proton from indirect dimension. Please open the NMRView Attribute window to check them.

After setting all the parameters, click Run->Get started

2. Overview of spectra

This  scripts will open 3 windows. The main assignment window is as below. All the operations are performed in the first 15N/13C spectra. The second spectra are used to display the slices that contain symmetry-related corsspeaks.

3. Symmetry-related crosspeak assignment

  1. Double click one crosspeak in .hsqc_2 or .chsqc_2 spectrum and you will get the corresponding slice in the main assignment window. Alternatively, you can use command, 'hsqcdsp/chsqcdsp'.
  2. Double click one NOE crosspeak and you will get a information window about its symmetry-related crosspeak as below;
  3. Error definition: error = 10.0*(pow(($xdif+$ydif)*20.0, 2.0)),  xdif, X axis difference;  ydif: y axis difference.
    Generally speaking, an acceptable error for one pair of symmetry-related corsspeaks within the same spectrum is small than 1.0. As for different spectra, it strongly depends on your spectrum calibration and an error of small than 2.0 is generally acceptable.
  4. Click the ASI button to assign the NOE. For the folded crosspeaks, please check the folding times beside the atoms.

4. NOE Assignment for non-symmetry-related crosspeak

  Locate the crosspeak and press "x" instead of double clicking it.For the folded crosspeaks, click the underline number "0", select the folding times and click NewAssi.

Please refer to the "Hotkeys" and "Commands" for more details
